Lipoprotein lipase (LPL), the main enzyme that controls plasma triglyceride concentrations, is a major determinant of triglyceride levels in the circulation. Alleles for two coding regions (S447X and S447X) in the LPL gene have been associated with plasma triglycerides (TG). However, the contribution of each of these variants to interindividual variation in plasma TG levels has not been established in large populations. We performed two genome-wide association studies of plasma LPL activity in up to 6,240 individuals of European ancestry. In the first analysis of the combined cohorts, the TG-raising allele was associated with lower LPL activity, and the opposite effect was seen for the TG-lowering allele. In the second analysis, the S447X single-nucleotide polymorphism was directly associated with LPL activity in separate cohorts of up to 4,769 individuals. However, the rs599839-T allele was associated with higher TG levels, an effect that was independent of LPL activity. The effects of both SNPs on TG levels were dependent on the plasma TG level at which they were measured. The S447X and LPL data are therefore complementary, with distinct effects of each SNP on LPL activity depending on the plasma TG level.PROJECT SUMMARY/ABSTRACT The African-American and Hispanic populations represent roughly 40% of the patient population in the Veterans Health Administration (VHA), and have the highest rates of diabetes, hypertension, heart disease, and other chronic conditions. Due to their demographics, these populations are also the most likely to use VHA services. By better understanding these populations' access to, and use of, VHA services, the VHA has a better chance of improving the overall quality of care for them.
